Characterization of lncRNA U90926 and its protein coding potential

Abstract
I am investigating the coding potential of novel mouse long non-coding RNA (lncRNA) U90926 (U9) based on off recent research that suggests lncRNAs may encode small but function proteins. U9’s open reading frame (ORF) contains a signal sequence that directs translation to the lumen of the ER. Through overexpression of tagged U9 ORF and immunocytochemistry, I determined that the U9 peptide is localized to the Golgi. Additionally, I confirmed that it follows this secretory pathway and is secreted via western blot. Current investigation is aimed at creating a monoclonal antibody to endogenously detect U9 and determining any post translational modifications.
