Enzyme Modification of Carbon Fiber Microelectrodes for in Vivo Acetylcholine Detection Using Fast-Scan Cyclic Voltammetry

Abstract
Abnormal release of acetylcholine (ACh) in the brain has been implicated in major neurological disorders like epilepsy, however few technologies have been developed to allow for rapid and spatially precise in vivo measurements of this neurotransmitter. We are optimizing an enzyme modification method on carbon fiber microelectrodes for sensitive and selective detection of ACh. Sensitivity and stability were characterized in a flow cell using fast-scan cyclic voltammetry. Significant events were recorded and quantified during preliminary in vivo testing. Future experiments will continue optimization and in vivo testing in rat hippocampi for eventual elucidation of the role of ACh in epilepsy.
