REV1 Depletion Sensitizes Colorectal Cancer Cells to Chemotherapeutics in an Autophagy Dependent Manner

Abstract
The current treatment modalities for colorectal cancer remain extremely limited due to resistance and frequent relapse. Colorectal cancer has emerged as the leader in cancer deaths below the age of 50, demonstrating the need for novel therapeutic strategies. Previously, inhibition of REV1 was shown to sensitize cancer cells to chemotherapeutics. In this study, inhibition of the REV1 translesion synthesis polymerase sensitized colorectal cancer cells to chemotherapeutics regardless of mismatch repair status, by a mechanism not dependent on DNA damage accumulation, rather by inducing autophagy and diminishing cystoskeletal protein expression. Thus, REV1 presents as a promising target to sensitize colorectal cancer cells to standard-of-care chemotherapeutics.
