Abstract
Raman spectroscopy, an inelastic light scattering technique, is a tool that can probe molecular vibrations of proteins. Raman spectroscopy can quantify a protein’s averaged Ramachandran φ-angles due to its relationship with the Amide III vibrational mode. However, a relationship between the Ramachandran φ-angle and a vibrational mode has not been determined. We developed a method to crystallize dipeptides with varying Ramachandran φ-angles to correlate the φ-angles to the Amide III vibrational mode. This relationship will be applied to enhance the structural characterization of amyloid fibril protein aggregates, which are implicated in Alzheimer’s disease.
