Determining the Role of the LATD/NIP Gene in Nodule Development in Lotus japonicus

Abstract
The LATD/NIP gene is required for lateral root and symbiotic nodule meristem function. Some legume species, like Medicago truncatula, maintain a meristem in their symbiotic nodules and some do not, like Lotus japonicus. To determine if the presence of a nodule meristem affects LATD/NIP expression, a construct containing the uidA reporter gene driven by the LjLATD/NIP promoter will be transformed into Medicago truncatula and Lotus japonicus. Additionally, a construct containing the uidA reporter driven by the MtLATD/NIP promoter will be transformed into Lotus japonicus to observe expression patterns of a meristem gene in a legume that lacks a persistent nodule meristem.
