Identification of 5-hydroxymethylcytosine markers across four tissues in the cattle brain

Abstract
We investigated the presence of 5-hmC in four limbic system tissues. WGBS and oxRRBS were used to identify 5-hmC across the genome at single nucleotide resolution. We identified 56,254 5-hmC sites in Cingulate gyrus (Ci), 47,369 sites in Prefrontal cortex (PFC), 3,499 sites in Periaqueductal gray (PAG), and 3,541 sites in Hippocampus (Hi). The presence of 5-hmC varied in a tissue-specific manner. Differential 5-hmC was identified between docile and non-docile animals. Both PFC and Hi had significant differential hydroxymethylation around the same location on chromosome 27, on Bos taurus gene RN18S1. These results show 5-hmC as a tissue-specific epigenetic factor.
