Abstract
The study focused on developing and tweaking a protocol for extracting high quality DNA from the sea urchin, Strongylocentrotus purpuratus, specifically from frozen tissues. The yields and quality of frozen individuals were compared to that of live individuals. Tissue from the frozen individuals were taken from the mouth, stomach, Aristotle's Lantern, tube feet, eggs, and sperm. Tissue from fresh individuals were taken from the tube feet and base of the spines. It was found that fresh tissue had much higher yields and quality than that of frozen tissue.
