Characterization of the Receptors that Mediate the Prokinetic Effects of 5-HT4 Agonists on Propulsive Motility

Abstract
Functional gastrointestinal disorders are very common, with a frequent symptom of these conditions being constipation. One of the most effective treatments for constipation involves 5- HT4 receptor agonists, but it is not clear whether they act on receptors in the mucosa or muscularis. Transgenic mouse strains in which this receptor is tissue-specifically down-regulated were utilized and motility assays were performed with test compounds that activate the 5-HT4 receptor. The agonists were found to have inconsistent results on the different mouse strains, which may be a result of ineffective knockdown of the receptors.
