Abstract
Functional gastrointestinal disorders are very common, with a frequent symptom of these conditions being constipation. One of the most effective treatments for constipation involves 5-HT4 receptor agonists, but it is unclear as to whether they act on receptors in the mucosa or muscularis. Motility assays were performed with test compounds that activate the 5-HT4 receptor on transgenic mouse strains in which this receptor is tissue-specifically downregulated. The agonists were found to have inconsistent results on the different mouse strains, which may be a result of ineffective knockdown of the receptors.
